摘要
目的通过构建traP突变菌株研究RNAⅢ激活蛋白靶蛋白(TRAP)蛋白在金黄色葡萄球菌(SAU)中的功能。方法利用同源重组的方法在金黄色葡萄球菌(SAU)8325-4中构建traP的缺失突变菌株,对其表型和几种压力条件下的生长和生存状况进行检测,揭示其在金黄色葡萄球菌生命活动中的作用。结果在金黄色葡萄球菌8325-4中成功构建了traP的缺失突变菌株,突变株生长缓慢,其他表型如疏水性、表面电荷和生物膜发生了明显的变化;在重金属、过氧化氢,以及全血杀伤等压力条件下的生长和生存状况也发生了明显改变。结论TRAP蛋白在金黄色葡萄球菌侵袭机体的过程中发挥着重要的功能。
Objective To study the function of target of RNAⅢ-activating protein(TRAP) in Staphylococcus aureus by constructing the traP mutant.Methods The traP deletion mutant was constructed by homologous recombination in S.aureus 8325-4 a series of assays was performed to check the alternation of the phenotypes and stress responses in order to reveal the function of TRAP.Results The traP mutant was successfully constructed in S.aureus 8325-4.Compared with its parent strain,the growth rate of the mutant was decreased and other phenotypes such as hydrophobicity,surface charge and biofilm were changed remarkably.The growth and survival under stress conditions,such as heavy metal,hydrogen peroxide,human bloD killing,were altered notably.Conclusion TRAP may play an important role in the pathogenicity of S.aureus.
