摘要
目的:探讨人工虫草复合液对大鼠离体肾缺血再灌注损伤的保护效应及其作用机制。方法:将72只雄性SD大鼠离体肾脏随机分为生理盐水灌注组、高渗枸橼酸盐腺嘌呤液(HCA液)灌注组和虫草复合液灌注组,分别观察灌注后6h、12h、24h肾损伤程度、肾小管上皮细胞凋亡指数(AI)、肾组织匀浆中超氧化物歧化酶(SOD)活力与丙二醛(MDA)含量。结果:(1)在灌注后6h,与生理盐水组和HCA液组比较,虫草复合液组肾损伤程度较轻;灌注后12h,与生理盐水组比较,HCA液和虫草复合液组肾损伤程度较轻,而HCA液和虫草复合液组肾损伤程度无差异;在灌注后24h,各组肾损伤程度无差异;(2)与HCA液和生理盐水组比较,虫草复合液组在灌注后6h、12h后AI较低(P<0.05)、肾组织匀浆中SOD活力较高(P<0.05)、MDA含量较低(P<0.05);灌注后24hAI、肾组织匀浆中SOD活力与MDA含量无统计学差异(P>0.05);(3)AI与肾组织匀浆中超氧化物歧化酶(SOD)活力负相关(r=-0.724),与丙二醛(MDA)含量正相关(r=0.681)。结论:人工虫草提取液灌注对大鼠离体肾缺血再灌注损伤具有一定的保护性效应,其作用机制可能与抗氧自由基、抗脂质过氧化作用有关。
Objective:To explore the protective effect and the mechanism of cordyceps extract on renal tissue in rats with ischemia reperfusion injury. Methodology: The kidneys of seventy-two male Sprague-Dawley rats were randomly divided into 3 groups: saline group, kidney preservation solution (HCA group) , and artificial cordyceps liquid composite group (CS-HCA group). We observed the degree of tubulo-interstitial lesions, apoptosis index (AI) of the renal tubular epithelial cell and superoxide disnmtase (SOD) activity and concentration of malondialdehyde aldehyde (MDA) in kidney at 6h, 12h and 24h after perfusion respectively. Results: ( 1 ) The degree of renal lesion in CS-HCA group was lighter than those in the Qther two groups at 6h 'after perfusion. The degree of renal lesion in both HCA group and CS-HCA group was lighter than saline group at 12h after perfusion. It was no differences for degree of renal lesion among 3 groups at 24h after perfusion. (2) Comparison with HCA group and saline group, CS-HCA group had lower AI( P 〈 0.05 )and MDA concentration ( P 〈 0. 05 ), and higher SOD activity( P 〈 0. 05 ) at 6 and 12h after perfusion. But no difference among three groups in AI and SOD activity, and MDA concentration (P 〉 0.05). (3) It was negative correlated between AI and SOD activity ( r = - 0. 724), positive ratio between AI and MDA concentration( r = 0. 681 ). Conclusion: Cordyceps artificial compound could protect the kidney against renal ischemia reperfusion injury in rats, which of mechanism may be associated with the effects of free radical and lipid peroxidation.
