XAF1基因及蛋白在局灶性小肠胃肠道间质瘤组织中表达的研究 被引量:2

摘要 目的:探讨XAF1基因及蛋白在小肠胃肠道间质瘤(GISTs)组织中的表达及其与肿瘤危险度分级的关系。方法:分别采用实时定量聚合酶链反应(PCR)和蛋白印迹法检测XAF1在10例原发性局灶性小肠GISTs的肿瘤及瘤旁新鲜组织标本中的表达。结果:XAF1mRNA及其蛋白在小肠GISTs组织中的表达量显著低于瘤旁组织(P均<0.01)。中、高危小肠GISTs肿瘤组织中XAF1mRNA的表达均显著低于相应的瘤旁组织(P<0.05),而低危小肠GISTs与其瘤旁组织间XAF1mRNA的表达差异无统计学意义。结论:与瘤旁组织相比,XAF1基因的表达减少可能与小肠GISTs的发生有关。 Objective To investigate the expression of XIAP-associated factor 1 (XAF1) gene and protein in patients with gastrointestinal stromal tumors (GISTs) arising from small intestine and its relationship with tumor risk stratification. Methods Ten matched pairs of small intestine GISTs tumor tissue and para-neoplastic tissue were studied. The mRNA expression of XAF1 was determined by real-time PCR and the expression of XAF1 protein was determined by Western blot. Results The mRNA and protein expressions of XAF1 were significantly lower in GISTs tumor tissue than in paraneoplastic tissue (P0.05). The expression of XAF1 mRNA in GISTs tumor tissue was significantly lower than that in paraneoplastic tissue in moderate and high risk small intestine GISTs patients (P0.05), while no statistical significant difference in XAF1 mRNA expression was found in low risk small intestine GISTs patients. Conclusions The lowering of expression of XAF1 might be correlated with the development of GISTs.
