三种不同因子对三角帆蚌外套膜细胞Ca^(2+)流动性的影响 被引量:4

The Effect of Three Different Factors on Extracellular Ca^(2+) Flux of Mantle Cell from Hyriopsis cumingii

摘要 三角帆蚌(Hyriopsis cumingii)是我国主要的育珠母蚌,其所培育出的珍珠色泽鲜艳、细腻光滑,因而成为目前淡水育珠生产中首选的育珠母蚌之一。蚌的外套膜是贝壳和珍珠形成的重要组织器官,有内外两层表皮细胞及其间的结缔组织构成。它对钙具有高度通透性,其上皮细胞具有通过细胞膜主动吸收Ca(2+)和贮存Ca(2+)的功能,并通过胞吐作用排出钙至外套膜外腔中,这些钙就是形成珍珠的基础。 In order to study the molecular mechanisms of calcium metabolism in mantle cell from Hyriopsis cumingii,Ca(2+) flux direction and rate were detected by non-invasive micro-test technology(NMT).Three mantle cell cultures were divided into three groups(3 reduplication/each group).For the first group,the different concentrations of Ca^(2+)(0.5 mmol/L,1.25 mmol/L,3 mmol/L) were added to culture medium;for the second group,the different concentrations of vitamin D_3(control,50U/L,100U/L,500 U/L) were added into culture medium;for the third group,cells and tissues were cultured in the different temperatures(20℃,26℃,30℃),respectively.The results showed that extracellular Ca^(2+) flux was gradual changed from efflux to influx,and movement rate increased when adding Ca^(2+) into culture medium.The Ca^(2+) flux of 3 mmol/L Ca^(2+) adding group had significant difference compared with the control group(P0.01).The movement of Ca(2+) was gradual changed from efflux to influx,and movement rate increased when adding vitamin D_3 in cultured cells.Among of this,the Ca(2+) flux of 500 U/L vitamin D_3 group had significant difference compared with the control group(P0.01).Furthermore,culture temperature significantly affected the extracellular Ca^(2+) flux of the mantle cells.The flow directions of extracellular Ca^(2+)changed from efflux to influx with increasing temperature(P0.05).The influx rate at 26℃was the biggest among all the groups(P0.05).This experiment has a great significance on clarifing the formation and formation mechanism of pearls.
