摘要
目的通过对维吾尔族肥胖患者及3T3-L1脂肪细胞内脂素mRNA表达的检测,探讨内脂素mRNA表达与肥胖之间的关系。方法根据体重指数(BMI)将116名研究对象分为肥胖组和非肥胖组。①测量研究对象的体征指标和代谢参数,计算BMI和腰臀围比(WHR);②采用3-异丁基-1甲基黄嘌呤、胰岛素、地塞米松联合方案诱导3T3-L1前脂肪细胞分化为成熟的脂肪细胞;③应用RT-PCR方法检测受试者皮下和网膜脂肪组织以及3T3-L1前脂肪细胞在不同分化时段内脂素mRNA的表达水平。结果①肥胖组皮下脂肪组织内脂素的表达较非肥胖组显著升高(P=0.048),网膜脂肪组织内脂素的表达水平较非肥胖组有升高趋势,但差异无统计学意义。②相关性分析显示,网膜脂肪组织内脂素mRNA的表达量与BMI(r=0.26,P=0.047)、甘油三脂(TG)(r=0.298,P=0.000)和腰围呈正相关(r=0.191,P=0.041);皮下脂肪组织内脂素mRNA表达量与TG呈显著正相关(r=0.384,P=0.000)。③在3T3-L1前脂肪细胞的分化过程中内脂素mRNA表达水平逐渐升高,至分化末期达到较高水平且趋于稳定。其中,诱导分化后第6~12天较分化第0天显著升高(P<0.05),诱导分化第10~12天较第2~6天显著升高,诱导分化第12天较第6~8天显著升高。结论内脂素可能与维吾尔族肥胖及脂代谢有关;体外实验证实内脂素与肥胖的发生、发展有密切的关系。
Objective To explore the expression of visfatin mRNA in 3T3-L1 adipocytes in Uygur obese population and its association with obesity. Methods Totally 116 subjects were divided into obesity group and nonobesity group according to body mass index (BMI). (1) Measurement of anthropometric and metabolic parameters in the Uygur subjects and calculation of their BMI and wrist-hip ratio (WHR). (2) The differentiation of 3T3-L1 pre- adipocyte was induced by incubating the cells with 3-isobutyl-l-methylxanthine, insulin and dexamethasane. (3) RT- PCR was used to detect the expression of visfatin mRNA from subcutaneous, omental adipose tissues and mature 3T3-L1 adipocyte. Results (1) The expression of visfatin mRNA in subcutaneous adipose tissues in the obese group was significantly higher than that in the control one (P = 0. 048). (2) Correlation analysis showed that the expression of visfatin mRNA in omental adipose tissues was correlated with BMI ( r = 0.26, P = 0. 047), TG ( r = 0. 298, P = 0. 000) and waist circumference ( r = 0. 191, P = 0. 041). Visfatin mRNA expression in subcutaneous tissues was positively correlated with TG (r = 0. 384, P =0. 000). (3)During the differentiation phase of 3T3-L1 adipocytes, the expression of visfatin mRNA was increased, and the mRNA expression of visfatin gene remained a higher level and unchanged in the telophase. The expression level of visfatin mRNA from day 6 to 12 was higher than day 0 (P〈 0.05); from day 10 to 12 the expression of visfatin was higher than from day 0 to 6; the twelfth-day expression of visfatin was higher than from that from day 6 to 8. Conclusion Visfatin may be related to the development of obesity and lipid metabolism in Uygur population. Visfatin may have a close association with the occurrence and development of obesity.
