褪黑素对冈田酸致NG108-15细胞超微结构变化的作用

摘要 目的观察褪黑素(MLT)对冈田酸(okadaic acid,OA)致NG108-15细胞超微结构的影响。方法以光学显微镜、MTT法和电子显微镜观察OA对细胞形态和超微结构的影响及褪黑素的拮抗作用。结果 OA作用后细胞形态改变明显,突起回缩,细胞活性明显减弱,细胞空泡化明显,微管等结构松解,褪黑素组细胞活性显著提高,微管结构得到恢复。结论 MLT可对抗OA致阿尔茨海默病(AD)样细胞形态改变,可能与其对微管结构的保护作用有关,并且可能是其发挥抗AD的机制之一。 Objective To explore effect of melatonin on the change of NG108-15 cell ultrastructure induced by okadaic acid (OA).Methods OA was used to induce the Alzheimer's disease (AD) cell model in NG108-15.MTT method was used to measure cell viability.Optical microscope and electron microscopy were used to measure the morphology change of NG108-15 cell.Results OA could decrease the viability of NG108-15 cell in time and dose-dependent manner,and damage the cell's microtubule,but melatonin could protect the cell from OA.Conclusions Melatonin has an antagonistic effect on AD cell model and this maybe contributes to its effect on the cell's microtubule.
