摘要
目的探讨形觉剥夺性近视前后乙酰肝素酶(heparanase,HPSE)mRNA和基质金属蛋白酶(matrix metalloproteinase,MMP)-2mRNA在大鼠后极部巩膜中表达的时间动态变化。方法采用出生后2~3周的Wistar大鼠30只,随机分成A、B、C3组,每组10只,右眼分别行单眼眼睑缝合20d、40d、60d,左眼开放作为对照眼。于实验前、后剪取包括视盘在内的约3mm×3mm大小的后极部巩膜,行RT-PCR反应检测HPSEmRNA及MMP-2mRNA的表达。结果正常眼后极部巩膜均有HPSE mRNA和MMP-2mRNA少量表达。形觉剥夺前,HPSEmRNA和MMP-2mRNA表达组间及组内左、右眼比较,差异均无统计学意义(均为P〉005);形觉剥夺后,各组实验眼HPSEmRNA和MMP-2mRNA表达均较对照眼增加,差异均有统计学意义(均为P〈0.05)。结论大鼠在形觉剥夺诱导后实验眼后极部巩膜HPSE mRNA和MMP-2mRNA表达显著。
Objective To investigate the time dynamic changes of heparitinase(HPSE)mRNA and matrix metalloproteinase-2(MMP-2)mRNA expression in the posterior sclera of rats with form-deprived myopia.Methods Thirty Wistar rats from 2 weeks to 3 weeks after birth were randomly divided into group A,B and C,10 cases in each group.The right eyes were monocular sutured of 20 days,40 days and 60 days,and the left eyes without operation were set as control group.The posterior sclera including the optic disc was selected with the area of 3 mm ×3 mm before and after the operation,the expression of HPSE mRNA and MMP-2 mRNA were detected by RT-PCR.Results A little of HPSE mRNA and MMP-2 mRNA were expressed in the posterior sclera of normal eyes.Before form-deprived,there was no statistical difference in HPSE mRNA and MMP-2 mRNA expression compared between the right and left eyes(all P0.05).After form-deprived,the HPSE mRNA and MMP-2 mRNA expression in the right eye of each group were higher than that in the left eyes,there were all statistical differences(all P0.05).Conclusion The expressions of HPSE mRNA and MMP-2 mRNA in the posterior sclera of rats with form-deprivation are significant.
