摘要
目的研究中国浙江汉族成年男性载脂蛋白C-1(Apo C-1)基因型与血浆脂蛋白相关磷脂酶A2(Lp-PLA2)活性以及血脂代谢指标的关系。方法应用高分辨熔解(HRM)技术检测756例成年男性体检者Apo C-1rs4420638单核苷酸多态性(SNP)的基因型频率分布并检测血浆Lp-PLA2活性。同时检测三酰甘油(TG)、总胆固醇(TC)、低密度脂蛋白胆固醇(LDL-C)、高密度脂蛋白胆固醇(HDL-C)、血糖(Glu)、体重指数(BMI)和血压。结果 HRM可以清楚地区分Apo C-1基因rs4420638位点的AA型和AG型,未能检测到GG基因型。该SNP位点的AA和AG基因型在成年男性人群分布频率分别是76.1%和23.9%。不同的Apo C-1基因型个体中血浆Lp-PLA2活性差异有统计学意义。AG基因型个体的平均Lp-PLA2活性水平和LDL-C水平均显著高于AA型基因型携带者。结论 Apo C-1基因rs4420638 SNP位点的基因型可影响Lp-PLA2酶活性和LDL-C的水平。
Objective To study the relationship between genotypes of apolipoprotein C-1(Apo C-1) and the activities of lipoprotein-associated phospholipase A2(Lp-PLA2) as well as the parameters in the blood lipid metabolism of adult male Han Chinese in Zhejiang.Methods High resolution melting(HRM) was used to detect the single nucleotide polymorphism(SNP) of Apo C-1 rs4420638 in 756 adult male Han Chinese subjects,and the activities of Lp-PLA2 were determined as well.The levels of triglyceride(TG),total cholesterol(TC),low-density lipoprotein cholesterol(LDL-C),high-density lipoprotein cholesterol(HDL-C),glucose(Glu),body mass index(BMI) and blood pressure were determined.Results AA and AG genotypes of Apo C-1 rs4420638 were unambiguously distinguished by HRM.The occurrence of GG genotype was completely absent.The prevalences of the AA and AG genotypes for its SNP site were 76.1% and 23.9% respectively.There were significant differences in the activities of Lp-PLA2 between two groups with different genotypes.However,the average levels of Lp-PLA2 activity and LDL-C in individuals with AG genotype were significantly higher than those with the AA genotype.Conclusions The genotype of Apo C-1 rs4420638 may contribute to the variation of Lp-PLA2 activity and the level of LDL-C.
