荞精补肾胶囊对糖尿病大鼠心肌病变的影响及机制

Effects and mechanism of Qiaojing Bushen capsule (QJBS) on cardiomyopathy in rats with diabetes mellitus

目的观察荞精补肾胶囊对实验性糖尿病大鼠心肌病变的影响并探讨其可能机制。方法用链脲佐菌素(STZ)和高脂饲料法诱导建立SD大鼠糖尿病模型,将造模成功的53只大鼠随机分成4组,荞精补肾胶囊高剂量组(高剂量组)、低剂量组各14只,分别灌服蒸馏水配制的荞精补肾胶囊溶液1.2、0.6 g(0.3 ml)/kg;消渴丸组12只,灌服蒸馏水配制的消渴丸溶液0.83 g(0.3 ml)/kg;模型对照组13只,灌服蒸馏水0.3 ml/kg;以上各组继续以高脂饲料喂养;正常对照组12只,灌服蒸馏水0.3 ml/kg,继续以普通饲料喂养。5组均每天灌胃1次,持续8周。给药结束后测空腹血糖(FBG)、TC、TG、游离脂肪酸(FFA)、心重指数(HWI)、血浆TNF-α水平,光镜下观察心肌病理变化,同时用电子显微镜观察心肌超微结构变化,并采用免疫组化法检测心肌组织中TNF-α蛋白表达。结果与模型对照组比较,高、低剂量组FBG、TC、TG、HWI水平及血浆中TNF-α表达均明显降低;光镜和电镜结果均显示心肌病理改变明显减轻。结论荞精补肾胶囊可抑制糖尿病大鼠心肌病理改变,对心肌起保护作用,机制可能为降低FBG、TC、TG、FFA水平及抑制TNF-α表达。

Objective To investigate the effects of QJBS on experimental diabetic cardiomyopathy in rats and its possible mechanism.Methods The rat model of diabetic cardiomyopathy was established by eating a high-fat diet together with injection of streptozocim（STZ）.Fifty-three diabetic cardiomyopathy rats were randomly divided into four groups,the QJBS high dose group（high dose group） and low dose group（low dose group） involved 14 in each,and were given QJBS 1.2,0.6 g（0.3 ml）/kg（solved with distilled water）;the Xiaokewan group（n=12）was given Xiaokewan 0.83 g（0.3 ml）/kg（solved with distilled water）;the model control group（n=13）was given distilled water 0.3 ml/kg.All the aboved groups were given high-fat diet.The normal control group（n=12） was given distilled water 0.3 ml/kg and normal diet.The five groups all administered the treatment intragastrically once a day for 8 weeks.At the end of the experiment,fasting blood glucose（FBG）,total cholesterol（TC）,triglyeride（TG）,free fatty aids（FFA）,the heart weight index（HWI） and TNF-α were measured.The cardiac muscle ultrastrueture was observed.Immunohistochemical technique was used to detect TNF-α expression.Results Compared with model control group,FBG,TC,TG,FFA,HWI decreased markedly in the high dose group and low dose group;light microscope and electron microscope revealed that the pathological changes lightened.Conclusion The QJBS can inhibit the pathological changes of diabetic cardiomyopathy,and has protective effects on cardiac muscle in rats of diabetes Mellitus.The mechanism may be related with inhibition of FBG,TC,TG,FFA and TNF-α level.
