摘要
目的探讨大面积急性肺血栓栓塞(AMPE)家兔心肌受损情况及可能机理。方法模型组10只家兔静注血栓栓子制作AMPE模型;假手术组10只家兔造模过程中只注射生理盐水;对照组10只家兔不做任何处理。于造模成功后2、24 h采集三组静脉血,分别采用微粒子化学发光法、流式细胞仪法、放射免疫法、酶联免疫双抗体夹心法测定血浆心肌肌钙蛋白(CTnI)、血小板膜表面糖蛋白(GPⅡb/Ⅲa)、内皮素-1(ET-1)及血管性假血友病因子(vWF)、α-颗粒蛋白(GMP-140)水平。结果模型组2、24 h血浆CTnI、vWF、GPⅡb/Ⅲa、ET-1、GMP-140水平均明显高于假手术组(P均<0.05)。结论 AMPE可致心肌受损,机制可能与血浆vWF、GPⅡb/Ⅲa、ET-1、GMP-140水平升高有关。
Objective To investigate the myocardial damages and its mechanism in acute massive pulmonary embolism(AMPE)rabbits.Methods The model group with 10 rabbits was made AMPE model by intravenous injection of thromboembolus;the sham group with 10 rabbits was only injected physiological saline;the control group with 10 rabbits were not intervened.2,24 hs after the successful preparation of model,the venous blood of rabbits were collected,the plasma level of cardiac troponin(CTnI),platelet membrane surface glycoprotein(GPⅡb/Ⅲa),endothelin-1(ET-1) and vascular false hemophilia factor(vWF),alpha particle protein concentration(GMP-140) were detected respectively by means of micro particle chemiluminescence method,flow cytometric analysis method,radioimunoassay method,enzyme-linked immunosorbent double antibody clip neo-confucianism.Results The 2,24 hs plasma level of CTnI,vWF,GPⅡb/Ⅲa,ET-1,GMP-140 were significantly higher than those in the sham group(all P0.05).Conclusion AMPE can induce myocardial damage,the mechanism maybe related to the increasing of vWF,GPⅡb/Ⅲa,ET-1,GMP-140 level.
