摘要
胚胎干细胞(ESCs)是遗传工程、发育生物学和疾病模型研究与应用的重要材料。猪ESCs建系研究虽已历经十多年,且已有建立猪类ES细胞系的报道,但由于其培养和鉴定方法基本都是借鉴小鼠和人的ESCs建系,从而使其培养体系存在着极大的不可靠性,至今未有种系嵌合的猪ESCs建系成功的报道。论文对猪ESCs建系的影响因素和存在的问题、传统的培养和鉴定方法在猪ESCs建系中的应用、ESCs多能性分子以及ESCs研究中出现的新理论进行了综述。
Embryonic stem cells(ESCs) are the essential materials in the research and application of genetic engineering,developmental biology and disease model.Research on porcine ESCs has gone through more than a decade,and the establishment of ES like cell lines has been reported,but due to the current methods of cultivation and identification of porcine ESCs draw lessons from mouse's and human's ESCs lines establishment,the culture system is so unreliable that no report of obtaining porcine germ-line chimeric ESCs lines has been confirmed yet.In this article,the factors influencing procine ESCs cell lines establishment and research problems,the appliation of the traditional cultivation and identification methods in porcine ESCs research,ESCs multipotential molecules and new theories in this scope were reviewed.
