摘要
[目的]探讨壳聚糖诱导子对喜树悬浮培养细胞喜树碱积累的影响。[方法]在壳聚糖诱导的喜树悬浮培养体系中,测定培养细胞的干重、喜树碱的产量、过氧化氢和丙二醛的含量。[结果]在喜树悬浮培养细胞第20天加入最终浓度80mg/L的壳聚糖诱导子刺激36h后,喜树碱的总产量达到15.21mg/L,是未作处理的喜树碱总产量最大值(在细胞培养的第20天达到)的6.18倍。同时,受到诱导的培养细胞内过氧化氢和丙二醛的含量都急剧增加。[结论]壳聚糖诱导子的刺激能显著地促进喜树悬浮培养细胞喜树碱的生物合成。
[Objective] The aim is to study the role of the chitosan elicitor in camptothecin accumulation with suspension cultures of Campototheca acuminata Decaisne.[Method] Cell dry weight,camptothecin yield,hydrogen peroxide(H2O2)and malondialdehyde(MDA)content were investigated in suspension cultures of C.acumunata induced by the chitosan elicitor.[Result] When the final concentration of 80 mg/L elicitor was added to the suspension cultures on its 20th day of growth,the highest camptothecin yields(15.21 mg/L)was achieved after 36 hours' subcultures,6.18 times that of the untreated cells,which obtain their maximum yield at the 20th day of culture.Correspondingly,higher hydrogen peroxide and malondialdehyde content occurred in elicited cells during subsequent cell suspension subcultures.[Conclusion] Chitosan elicitor could significantly promoted camptothecin biosynthesis with cell suspension cultures of C.acuminata.
