摘要
目的 研究锌缺乏影响机体免疫功能的机理以及与细胞凋亡的关系。 方法 采用DNA 琼脂糖电泳及流式细胞仪分析(FACS) ,观察了不同浓度的硫酸锌对缺乏营养因子引起的胸腺细胞凋亡的影响。 结果 随培养液中硫酸锌浓度的增加,细胞死亡率逐渐增加;DNA 琼脂糖电泳显示,103 ~105 浓度的硫酸锌可抑制胸腺细胞自发性的凋亡;FACS 分析表明,凋亡的抑制率可达60 % ~65 % 。 结论 锌可抑制胸腺细胞凋亡,促进细胞免疫功能,对营养缺乏导致的免疫功能低下具有抑制作用。
Objective To observe zinc sulfate on the effcets of thymoytes apotosis induced by removing growth factore. Methodes Apoptosis was measured by electron microscope.DNA electrophoresis and flow cytometric methods. Results Thymocytes apoptosis happened when cultured with serum free medium for 20 hrs,appering apototic bodies and DNA ladder.Zinc sulfate could inhibit this apoptosis at a concentration from 10 5 ~10 8 mol/L,and a dose dependent effect was observed,higher than 10 3 M could incerase cell necrosis. Conclusion Zinc sulfate can inhibit serum free induced thymocytes apoptosis,increase cellular immune function,and plays an important role in prevention and treatment of infectious deseases.This mechanism requires for further research.
