摘要
目的探讨妊娠对兔骨折愈合的影响。方法将70只日本大耳白雌兔按随机数字表法分成对照组(n=30)、妊娠前组(n=40),对照组不做处理。妊娠前组交配受孕,受孕后选出30只组成妊娠组。妊娠组孕后9d分组造模成左小腿中段闭合骨折模型,按造模前,造模后即时,造模后1、2、3周各时相点抽取耳缘静脉血或心穿血,提取血清后-80℃冻存,一并送检生长激素,按时组织学检查,免疫组化检查、计算机数字化X线成像(computed radiography,CR)检查,其数据行统计学检验。结果妊娠组造模后即时,造模后1、2、3周血清生长激素值[(1.331±0.067)、(1.426±0.024)、(1.507±0.109)、(1.826±0.289)ng/ml]高于对照组[(1.208±0.151)、(1.317±0.102)、(1.246±0.192)、(1.340±0.313)ng/ml](P<0.05);妊娠组造模后1、2、3周的骨形态发生蛋白(bone morphogenetic protein,BMP)平均光密度值和平均积分光密度值均高于对照组(P<0.05),CR定量测得的骨痂平均光密度值均高于对照组(P<0.05)。结论兔早中期妊娠可能对骨折愈合有促进作用。
Objective To investigate the effect of pregnancy on fracture healing of rabbit. Methods A total of 30 pregnant Japanese big ears white female rabbits and 30 matched female rabbits were subjected. Fracture model was established in 9 d after pregnancy. Before, and 1, 2, and 3 weeks after model establishment, their blood was extracted and stored at -80 ℃ for growth hormone with radioimmunoassay. Callus samples were collected for morphological and immunohistochemical study. Computed radiography (CR) was performed for radiological data. Results Serum growth hormone in pregnant group was 1.331±0.067, 1.426±0.024, 1.507±0.109 and 1.826±0.289 ng/ml respectively in 0, 1, 2 and 3 weeks of model establishment, significantly higher than those in control group except for the time of model establishment (1.208±0.151, P0.05; 1.317±0.102, 1.246±0.192 and 1.340±0.313 ng/ml for the 3 time points, P0.05). There was more bone morphogenetic protein (BMP) expressed in the callus of pregnant group than in that of the control (P0.05). CR indicated that more condensed callus was observed in the pregnant group than in the control (P0.05). Conclusion Pregnancy in early metaphase promotes fracture healing in rabbits.
