摘要
通过对本实验室保藏的一株放射性土壤杆菌Agrobacterium cradiabacter发酵生产可得然胶时的氮源、碳源等培养基组分及发酵条件的研究,得出了菌株产可得然胶的最佳培养基配方及最适培养条件。结果表明,最佳培养基配方为:葡萄糖6.0%,(NH4)2HPO40.24%,KH2PO40.2%,MgSO4·7H2O0.1%,CaCO30.2%,营养因子浓缩液0.25%,pH7.0。最佳培养条件为:装液量80ml/500ml三角瓶,摇床转速为180rpm,培养温度30℃,发酵时间120hr。在此条件下,可得然胶产量达41.0g/L,糖胶转化率高达68.3%。
By the research of nitrogen source, carbon source, other medium components and curdlan fermentation conditions, the optimal medium was established as follows: glucose 6.0%, (NH4)2HPO4 0.24%, KH2PO4 0.2%, MgSO4·7H2O 0.1%, CaCO3 0.2%, neurotrophic factor concentrated liquid 0.25%. The optimal culture conditions were that: initial pH 7.0, volume 80ml of 500ml Shake flask, shaking speed 200 r / min, temperature 30℃ with this medium. After 120 hours fermentation, the highest yield of curdlan was 42.0 g/L and its conversion rate was 70%.
