摘要
目的探讨白血病抑制因子对体外培养神经干细胞增殖和分化的影响。方法观察体外培养神经干细胞在添加白血病抑制因子环境下的增殖、分化状况。结果体外培养神经干细胞呈悬浮生长,具有不断增殖的能力。添加白血病抑制因子能加快神经干细胞的增殖速度,提高其长期增殖能力。加血清诱导分化后细胞可分化为不同形态的细胞。添加白血病抑制因子可以抑制神经干细胞的分化,保留其多能型。结论白血病抑制因子对促进神经干细的增殖及维持未分化状态具有重要作用。
Objective To observe and analyse the effects of leukemia inhibitory factor(LIF) on proliferation and differentiation of neural stem cells(NSCs) in vitro. Methods LIF was used to induce NSCs in vitro. Results Nses cufhured vitro are suspended growth and have the ability to proliferate continuously. LIF can accelerate their protife sprrd ationand enhance their ability of long term proliferation. Fetal calf serum can induce, the differentiation at Nscs, but LIF inhibit the differentiation of NSCs and retain their multi-energy-type. Conclusion LIF can promote the proliferation and maintain non-differentiated form of NSCs.
