摘要
目的:观察缺氧诱导因子-1(hypoxia induciblefactory-1,HIF-1)以及其拮抗剂对人脑胶质瘤细胞株T98G放射线敏感性的影响,初步探讨缺氧影响放疗疗效的机制。方法:以化学性缺氧模型观察缺氧诱导因子-1对细胞放射敏感性的影响,以及缺氧诱导因子-1拮抗剂毛壳菌素的放射增敏作用。结果:氯化钴显著降低放射线对T98G细胞的增殖抑制作用,使用毛壳菌素拮抗缺氧诱导因子-1的转录活性可逆转放射线脱敏现象。结论:氯化钴抑制胶质瘤细胞的放射线敏感性,该作用与缺氧诱导因子-1的转录活性相关。
Objective: To observe the effect of hypoxia incucible factor-1 (HIF-1 ) on the sensitivity of T98G human glioma cells to radiotherapy and the abrogation of the effect by the antagonist of HIF- 1, hereby provide novel evidence to the radioresistant effect of HIF-1. Methods: Chemical hypoxia model was employed to induce HIF-1 in T98G glioma cells. Proliferation upon the irradiation and/or HIF-1 antagonist was observed by MTT assay. Results: Cobalt chloride desensitized the T98G glioma cells to the proliferation-inhibiting effect of irradiation while the HIF-1 -autagomst chetomin restored this inhibitory effect. Conclusion: HIF-1 plays an essential role in the radioresistance in T98G human glioma cells.
