摘要
目的:确定纯化江南卷柏总黄酮的大孔树脂及其工艺参数。方法:以总黄酮的含量为指标,从3种不同型号的大孔树脂中筛选纯化江南卷柏总黄酮的最佳树脂;以江南卷柏总黄酮的回收率为指标,通过单因素试验确定该树脂分离纯化的工艺条件。结果:D101型树脂对江南卷柏总黄酮有较好的吸附和解吸附性能;其纯化江南卷柏总黄酮的较佳工艺条件为:药材量与树脂量的比例为1∶3,以8BV(8倍体积)70%乙醇洗脱,流速为2BV·h-1,树脂每次可重复使用3次。在所确定的工艺条件下,江南卷柏总黄酮纯度可达50%以上,回收率可达85%以上。结论:该工艺条件可用于江南卷柏总黄酮的纯化。
Objective:To determine the optimum macroporous resin the parameters of technology purificating total flavonoids from Selaginella moellendorfii Hieron with the resin.Methods:With the content of total flavonoids from Selaginella moellendorfii Hieron as index,screen out the optimal resin which can purify the from 3 different resins,and determin the technology condition purifying the by single-factor experiments.Results:The total flavonoids from Selaginella moellendorfii on D101 macroporous resin showed the best effection of adsorption desorption.The preferable technology of purifying the total flavonoids was:the ratio of weight of the herbs and resin was 1∶3,eluted at a velocity of 2 BV·h^-1 by a volume of 8 BV·h^-1 of 70% alcohol.The resin could be reused for 3 times before treating.Under the technology the recovery rate of total flavonoids was more than 85% and the purity was more than 50%.Conclusion:The D101 macroporous resin is suitable to purify the total flavonoids from Selaginella moellendorfii Hieron.
