红花醇沉液浓缩除醇过程中多元质控指标的近红外快速检测 被引量:10

摘要 目的:应用近红外光谱法,实现中药浓缩除醇过程中多元质控指标的快速检测,实时反映浓缩过程的状态,真正地实现浓缩过程的质量控制。方法:本研究以红花醇沉液浓缩除醇过程为例,收集浓缩过程样本和采用浸膏配制不同乙醇浓度的样本作为校正集,建立红花浓缩过程羟基红花黄色素A(Hydroxysafflor yellow A,HSYA)浓度、溶液密度的近红外校正模型,并对未参与建模的浓缩过程样本进行预测。结果:所建立的HSYA、密度模型相关系数R均达到0.990,采用独立的验证集对模型进行验证,预测集相关系数R2均大于0.980,RSEP小于10%。同时,本研究还拟合出红花乙醇溶液密度与醇度的关系曲线,其相关系数R2达到0.998。通过测得的密度值,带入本研究所建拟合方程即可获得样本实际乙醇浓度值,可有效地避免了气相色谱测定的烦琐操作,缩短分析周期。结论:研究结果表明,利用NIRS分析技术可以实现红花醇沉液浓缩过程中HSYA浓度、密度、乙醇浓度三个关键质控指标参数的同时快速检测。 Objective:To really realize quality control of Chinese herb concentrating process, near-infrared spectroscopy (NIRS) was used for fast monitoring multiple quality-control indexes of the process simultaneously, which could real-timely reflect its state.Methods: Taking the concentrating process of Carthamus tinctorius L.(CT)alcohol sedimentation solution as an example, samples were composed of two parts, one collected in the process and another prepared by diluting concentrated CT alcohol sedimentation solution.Models of HSYA content and density were established based on PLS algorithm, which were applied to predict the samples in the process of concentration which didn't participate in modeling to test the performance of the models.Results: The correlation coefficients of both calibration models achieved 0.990.And the predicted coefficients were up to 0.980, RSEP for them were less than 10%.Moreover, the fitting curve of the density and alcohol concentration were developed, the correlation coefficients of which reached 0.998.Therefore, the alcohol concentration could be quickly attained by the curve with known density so that the complicated operations of gas chromatography were avoided and analytical time was cut.Conclusion: It is concluded that NIRS can be used as a rapid method to determinate three key quality control indexes (the density, concentration of HSYA and ethanol) at one time in the process of concentrating process of Carthamus tinctorius L.alcohol sedimentation solution.
