摘要
以拟南芥为材料介绍一种不需组培的原位植物转化方法—真空渗透法。将含有T—DNA载体的农杆菌细胞悬液,用真空渗透的方式转化愈伤的完整植株,从而直接获得转化的种子。该法快速简便、重复性好,不需经过组培阶段即可获得转化植株。

transformation, vaccum infiltration is presented. The approach is vaccum infiltration of a
suspension of Agrobacterium cells containing T-DNA vector into Arabidopsis thaliana; thereby
the transformed seeds can be harvested directly. This reproducible, non-tissue cultural
procedure yields stably transformed A. thaliana seeds and its transformation frequency is high
enough for introducing foreign genes and characterizing their expression.
