摘要 本实验利用神经细胞培养技术,用加入NGF的正调法和加入NGF抗体、NGF受体抗体的负调法、观察了NGF及其受体在胚胎神经细胞发育和存活中的作用。结果显示:NGF可明显促进神经细胞突起生长和细胞存活.加入NGF抗体和NGF受体抗体则引起神经细胞死亡。在培养的第3d.NGF受体开始表达.7d后达到高峰,而后逐渐减少,结果表明:NGF及其受体在胚胎神经细胞发育中有着非常重要的作用。 Using primary cultures of neural cells with addition of nerve growth factor (NGF),anti-NGF and anti-NGFR (trkA)antibodies, we further observed the biologic effects of NGF and its receptor on the development and survival of embryonic neuralcells. The results showed thai NGF was increased both in the number of survival cells and in the outgrowth of neurites. Additionof anti-trkA and anti NGF antibodies induced apoptosis of neural cells. The expression of trkA on neural cells began to occurfrom 3 day cultures. reached its climax in 7 day culture, then gradually decrease. The results suggest that NGF and its receptorplay an important role in the development of embryonic neural cells.
