摘要
目的观察苦参碱抑制人肺腺癌细胞A549的增殖作用,并探讨其可能机制。方法采用XTT比色法观察A549细胞增殖、流式细胞术(FCM)检测细胞凋亡、免疫印迹检测凋亡相关蛋白表达水平、流式细胞术检测A549细胞F-actin的分布。结果苦参碱能明显抑制A549细胞的增殖,呈现剂量和时间依赖关系,能显著诱导A549细胞凋亡、降低凋亡相关蛋白Bcl–2表达(P<0.01),还可使A549细胞F–actin的含量增加。结论苦参碱可显著抑制A549细胞的增殖并诱导其凋亡,其机制可能与下调Bcl–2蛋白表达水平有关。
Objective To investigate the effects of matrine on proliferation of line A549 with lung adenocarcinoma cultured in vitro.Methods The A549 cells were treated with matrine in different concentrations and variable durations.The effect of matrine on A549 cells proliferation inhibition,apoptosis promotion,F-actin location and Bcl-2 protein expression were observed by the methods of cell growth curves,XTT,Flow cytometry and Western-blot.Results Compared with the control group,Matrine at the concentration of 0.1 to 0.5mg·mL-1 had a significant time and dose-dependent effect on proliferation inhibition(P0.01).FCM detection showed matrine could induce apoptosis and increase expression of F-actin in A549 cell.The expression of bcl-2 protein was significantly decreased in the matrine groups by Western-blotting.Conclusion Matrine can efficiently inhibit the proliferation and induce apoptosis of A549 cell,down-regulate bcl-2 protein expression which are involved in its anti-cancer effect.
