丹参酮ⅡA对心肌缺血再灌注BCL-2和Caspase-3表达的影响 被引量:7

摘要 为探讨丹参酮ⅡA(TanⅡA)对心肌缺血再灌注损伤大鼠Bcl-2及Caspase-3蛋白表达的影响,将大鼠随机分为假手术组、缺血再灌注组、TanⅡA低剂量治疗组和TanⅡA高剂量治疗组。用定量PCR方法观察缺血再灌注大鼠心肌Bcl-2和Caspase-3 mRNA表达,并采用末端脱氧核糖核苷酸转移酶介导的dUTP缺口末端标记法(TUNEL法)检测细胞凋亡。结果为缺血再灌注组Bcl-2和Caspase-3表达增加;与缺血再灌注组比较,TanⅡA高、低剂量治疗组均显著增加Bcl-2表达,减少Caspase-3表达。TanⅡA高、低剂量治疗组心肌梗死体积较缺血再灌注组减小。TanⅡA高、低剂量治疗组心肌细胞凋亡明显轻于缺血再灌注组,TanⅡA高剂量治疗组细胞凋亡亦轻于低剂量治疗组。TanⅡA对缺血再灌注心肌损伤能抑制其细胞凋亡,对心肌具有保护作用,其机制可能与增加Bcl-2蛋白表达同时降低Caspase-3蛋白表达有关。 Objective : To investigate the effect of tanshinone ⅡA on expression of BCL - 2 and caspase - 3 in rat with myocardial ischemia - reperfusion injury. Methods : The rats were randomly divided into pseudo - operation group, ischemia - reperfusion group, Tan ⅡA low - and high - dosage therapeutic group. Quantitative PCR was used to ob- serve the mRNA expression of Bcl - 2 and Caspase - 3 in myocardia of rat with ischemia - reperfusion, and TUNEL was used to detect the cell apoptosis. Results : Expression of Bcl - 2 and Caspase - 3 in ischemia - reperfusion group was increased compared with that of pseudo - operation group. While both groups of low - and high - dosage therapeutic Tan ⅡA, the expression of Bcl - 2 was significantly increased, and the expression of Caspase - 3 was downregulated. The volume of myocardial infarction was lower in High - and low - dosadge therapeutic Tan ⅡA compared to ischemia - reperfusion group. The apoptosis of myocardial cells in high - and low - dosadge group was less serious than that of ischemia - reperfusion group, and furthermore the eytoapoptosis in high - dosadge therapeutic Tan ⅡA group wao also less serious than that of low - dosadge therapeutic Tan ⅡA group. Conclusion : Tan ⅡA can inhibit cytoapoptosis in rat with myocardial ischemia -reperfusion injury, and be served to the protection of myocardia, which might be attributed to its function of increasing the expression of Bcl - 2 while decreasing the the expression of caspase -3.
