晚期非酶糖基化终末产物对大鼠甲状腺细胞甲状腺球蛋白及甲状腺过氧化物酶mRNA表达的影响 被引量:1

摘要 目的 研究晚期非酶糖基化终末产物(AGEs)对大鼠甲状腺细胞甲状腺球蛋白(TG)、甲状腺过氧化物酶(TPO)mRNA表达的影响,探讨糖尿病对甲状腺组织的损伤可能存在免疫机制以外的机制.方法 以FRTL细胞为研究对象,给予不同浓度AGEs刺激24、48、72 h后使用实时荧光定量PCR测定FRTL细胞TG mRNA和TPO mRNA的表达情况.结果 24 h:2.5 mg/L组(TG 2.27±0.25,TPO 2.5±0.17)、5.0 mg/L组(TG2.07±0.074,TPO 1.08±0.15)、10.0 mg/L组(TG 2.13±0.16,TPO 1.08±0.12)、AGEs抗体组(TG 2.48±0.12,TPO 2.43±0.89)表达水平均低于对照组(TG 5.64±0.16,TPO 5.19±0.25),差异有统计学意义(P<0.01);48 h:2.5 mg/L组(TG 1.29±0.89,TPO 3.44±0.40)、5.0 mg/L组(TG 1.22±0.16,TPO 3.17±0.21)、10.0 mg/L组(TG0.67±0.07,TPO1.4±0.13)、AGEs抗体组(TG 3.93±0.14,TPO 4.91±0.24)表达水平均低于对照组(TG 6.17±0.31,TPO 5.08±0.23),差异有统计学意义(P<0.01);72 h:2.5 mg/L组(TG 1.25±0.57,TPO 1.53±0.11)、5.0 mg/L组(TG 1.17±0.18,TPO 1.61±0.03)、10 mg/L组(TG 1.09±0.07,TPO 0.71±0.06)、AGEs抗体组(TG3.44±0.30,TPO 3.06±0.19)表达水平均低于对照组(TG 8.10±0.22,TPO 8.35±0.25),差异有统计学意义(P<0.01).结论 在体外从细胞和分子水平证明了AGEs对甲状腺细胞TG mRNA和TPO mRNA的表达水平有一定的影响,提示糖尿病对甲状腺组织的损伤可能存在免疫机制以外的机制.
