丹芍化纤对大鼠肺纤维化模型肺组织匀浆中SOD活性和MDA含量的影响 被引量:1

摘要 目的:察丹芍化纤对肺纤维化大鼠肺组织匀浆中超氧化物歧化酶(Superoxide Dismutase,SOD)、的活性和丙二醛(Maleic Dialdehyde,MDA)含量的影响。方法:SD大鼠30只,随机分为正常对照组(对照组)、肺纤维化模型组(模型组)和丹芍化纤胶囊治疗组(治疗组)。模型组和治疗组气管内注射博来霉素5 mg/kg诱导肺纤维化,对照组气管内注射等量生理盐水。次日起治疗组大鼠给予丹芍化纤混悬液0.8 g/(kg.d)灌胃,其余两组给予等量生理盐水灌胃。治疗第28 d处死各组大鼠,取肺组织行病理学观察、肺系数测定、肺组织羟脯氨酸含量、超氧化物歧化酶(SOD)活性及丙二醛(MDA)含量检测。结果:①与对照组比较,肺纤组大鼠肺系数、肺组织Hyp含量明显升高;肺泡炎症、肺纤维化程度明显;MDA含量明显升高,而SOD酶活性明显降低。②与模型组比较,丹芍化纤胶囊预防性治疗组大鼠肺系数、肺组织Hyp含量明显降低;肺泡炎症、肺纤维化程度均有所改善;MDA含量明显降低,但仍较正常组高;SOD酶活性明显回升,但未恢复正常水平。结论:丹芍化纤具有较好的抗大鼠肺纤维化作用,其作用机制部分是通过抗氧化作用抑制肺纤维化的发展,对肺纤维化有预防性治疗作用。 Objective: To observe the interventional effect of Danshaohuaxian (DSH) on the level of SOD and MDA from the lung tissues in rats with fibrosis pulmonary. Methods : Thirty Spague - Dawley rats were randomly divided into the control group, model group and treatment group. The model group and the treatment group were induced pulmonary fibrosis with bleomycin (5 mg/kg) endotracheally, while the control group was given with normal saline instead. From the second day of injection, the rats in the treatment group were administered intragastricly with DSH ( 0. 8 g/kg/d) . while the other rats were given normal saline instead. All rats were sacrificed on the twenty -eighth day, Histopathologicalhy examination, the lung coefficient, lung droxyproline (Hyp), SOD and MDA content was observed. Results: (1)Compared with NC group, it was revealed that the rats lung coefficient and lung tissue Hyp were significantly increased and the degree of pulmonary alveoli inflammation and pulmonary fibrosis was significantly induced in the PF group, rats lung tissue SOD were significantly decreased, and the MDA were significantly increased. (2)Compared with PF group, it was revealed that the rats lung coefficient and lung tissue Hyp were significantly decreased and the degree of pulmonary alveoli inflammation and pulmonary fibrosis was significantly relieved in the DSHC group, the rats lung tissue SOD were significantly increased and the MDA were significantly decreased. Conclusion: DSH has a good action against experimental pulmonary fibrosis which may be to elevate the activity of lung tissue SOD to alleviate oxidative stress.
