天津地区汉族人群E-选择素A561C基因多态性与2型糖尿病相关性研究

摘要 目的:探讨E-选择素基因A561C多态性与天津地区汉族人群2型糖尿病(T2DM)的关系。方法:利用试剂盒说明书方法提取DNA后,采用聚合酶链式反应-限制性片段长度多态性(PCR-RFLP)方法检测112例T2DM和健康体检者140例(对照组)A561C多态性,并采用微粒子化学发光全自动免疫分析法和全自动生化仪检测临床生化指标。结果:在112例T2DM患者中,检出12例杂合子突变基因型(AC),C等位基因频率为5.36%;在140例对照组中,检出4例AC,C等位基因频率为1.43%,2组均未检出突变纯合子(CC),差异有统计学意义(P<0.05);以基因型分组,112例T2DM患者中AC组空腹血糖(FPG)和餐后2h血糖(P2PG)值均高于AA组,空腹血胰岛素(FINS)低于AA组,差异有统计学意义(P<0.05)。结论:C等位基因可能为天津地区汉族人群T2DM易感基因,E-选择素基因多态性在T2DM的发生与发展中有一定作用。 Objective: To investigate the relationship between E-selectin gene A561C polymorphism and type 2 diabetes mellitus (T2DM). Methods:Using PCR-RFLP way, the A561C polymorphism was screened in 112 patients with T2DM and 140 controls. The clinical data were analyzed. Results:Twelve patients with the heterozygote mutation (AC) were found in 112 T2DM patients, and C allele frequency was 5.36%. Four were found in 140 controls, and C allele frequency was 1.43%. There was a significant difference between the two groups (P 0.05). No homozygote mutation (CC) was found in the two groups. The levels of fasting plasma glucose (FPG) and 2 h postprandial blood glucose (P2PG) were higher in T2DM patients of AC group than those of AA group. The level of fasting insulin(FINS) was significantly lower in T2DM patients of AC group than that of AA group (P 0.05). Conclusion: C allele may be the T2DM-susceptibility gene in Chinese Han population of Tianjin. E-selectin gene polymorphism plays an important role in occurrence and development of diabetes mellitus.
