参莲提取物对脐静脉内皮细胞功能的影响 被引量:2

Effect of Shenlian extracts on endothelial cells induced by TNF-α

摘要 目的:探讨参莲提取物对脐静脉内皮细胞(HUVEC)功能的影响。方法:建立TNF-α诱导内皮细胞炎性损伤体外模型,采用电镜法检测HUVEC形态学改变,渗透蛋白染色法检测细胞通透性,ELISA法检测细胞分泌功能,激光共聚焦显微镜检测细胞骨架F-actin和胞浆游离钙变化,观察参莲提取物对HUVEC功能的影响。结果:①参莲提取物各剂量组可显著改善HUVEC在炎症因子刺激下细胞结构的改变。②参莲提取物可降低渗透蛋白量,其中1mg/mL组差异有统计学意义(P<0.05)。③参莲提取物各剂量组可明显抑制HUVECF-actin高表达(P<0.05或P<0.01),并呈剂量依赖趋势。④参莲提取物各剂量组可显著抑制HUVEC胞浆游离钙异常增高(均P<0.01)。⑤参莲提取物对HUVEC分泌细胞因子未见明显作用。结论:参莲提取物可有效抑制炎性因子诱导下的内皮细胞结构和功能的改变,呈现出良好的抑制细胞炎性反应作用。 Objective:To investigate the effect of Shenlian(SL)extracts in preventing endothelial cells induced by TNF-α.Methods:Human umbilicus vein endothelial cell and TNF-αwere used to establish model for determining the permeability of endothelial cells in vitro,to observe morphology change by electron microscope,to observe HUVEC permeability function by protein staining method with transwell cabinet,and to observe the F-actin expression and active calcium concentration in cytosolic([Ca2+]ic)by confocal microscope,to observe effects on function cytokines secretion by ELISA.Results:①The results showed that model group’s lower layer protein level had been increased(P0.05).All SL groups had decreasing tendency,and 1mg/mL group’result was in evidence(P0.05).②All SL groups can inhibit HUVEC F-actin high express obviously,and the result showed that dose dependent trend(P0.01/P0.05).③SL extracts could reduce the elevation of[Ca2+]ic.④Model group’s IL-10 had decreased in evidence(P0.05),IL-6 had no observe changes.All SL groups had no obvious effect(P0.05).
