摘要
目的:观察三白草提取物对肥胖Zucker大鼠血糖及胰岛素抵抗的影响。方法:6周龄雄性肥胖Zucker大鼠12只,适应性喂养2周后,随机分为对照组和三白草提取物组(350mg/kg),所有动物给予高脂饲料喂养,疗程为4周。每周检测体质量、随机血糖;入组前、第14、28d时检测口服葡萄糖耐量试验(OGTT)和空腹血清胰岛素水平;第28d时检测血脂4项和血浆游离脂肪酸(FFA);第29d时,进行高胰岛素正葡萄糖钳夹试验,观察平均葡萄糖输注率(GIR)。结果:与对照组相比,治疗4周后,三白草提取物血清胰岛素轻度降低、高胰岛素正葡萄糖钳夹试验后GIR轻度升高;随机血糖、FFA均显著下降;体质量、进食量、OGTT有下降的趋势;对血脂无影响。结论:三白草提取物能轻度改善胰岛素抵抗。
Objective:To observe the effects of Saururus Chinensis on glucose metabolism and insulin resistance in obese Zucker rats.Methods:12 male obese Zucker rats,6 weeks old,were randomly divided into control group and Saururus Chinensis group(350mg/kg)after being fed for 2 weeks.All rats were feed with high fat diet and 4-week treatment.Body weight,food intake,and random blood glucose were tested every week.OGTT and fasting insulin level were tested on day 0,day 14 and day 28.TG,TC,HDL,LDL and FFA were tested on day 28.Glucose infusion rates were tested by hyperinsulinemic-euglycemic clamp from day 29.Results:Compared with the control group,the fed blood glucose had a significant decline in Saururus Chinensis-treated rats on day 28,even though there was no alteration of the fasting serum insulin and OGTT.The glucose infusion rates increased slightly in hyperinsulinemic-euglycemic clamp study.Furthermore,Body weight and FFA were decreased in Saururus Chinensis group.However,Saururus Chinensis had no significant effect on lipids in obese Zucker rats.Conclusion: Saururus Chinensis may improve glucose metabolism by reducing insulin resistance.
