大鼠气虚冻伤模型的建立及黄芪桂枝五物汤和桂枝汤作用研究

Establishment of qi deficiency-frostbite rats model and comparative effects and mechanism research of Huangqi Guizhiwuwu and Guizhi Decoction

目的:建立大鼠气虚冻伤模型并比较黄芪桂枝五物汤、桂枝汤对大鼠气虚冻伤模型的作用机制及相关程度高低。方法:采用气虚模型(过度疲劳+饮食失节法)复加冻伤模型(低温乙醇和水混合浸泡法)制备大鼠气虚冻伤模型,测定模型大鼠及黄芪桂枝五物汤、桂枝汤进行干预治疗后部分免疫细胞因子和TXB2、PGF1α、TXB2/PGF1α的变化。结果:与正常对照组相比,模型大鼠血清IL-1、IL-2、IL-6、TNF-β、TXB2、TXB2/PGF1α均显著升高(P<0.01或P<0.001),PGF1α显著降低(P<0.001)。与模型组相比,黄芪桂枝五物汤、桂枝汤均可显著降低IL-6、TNF、TXB2、TXB2/PGF1α的含量(P<0.05或P<0.01或P<0.001),显著升高PGF1α的含量(P<0.01或P<0.001),但两方对IL-2无显著性作用。黄芪桂枝五物汤还可显著降低IL-1的含量(P<0.01)。在对异常升高的IL-6、TXB2和TXB2/PGF1α的改善方面,黄芪桂枝五物汤组显著优于桂枝汤组(P<0.05)。结论:气虚冻伤模型造模3d存在炎症反应、免疫应激和血栓易形成状态,黄芪桂枝五物汤和桂枝汤均可抑制免疫应激和改善血循环障碍,使组织损害的病理过程减轻或改善。但黄芪桂枝五物汤在对病理损害的改善、抑制或减轻冻伤所致炎性反应及抑制血小板聚集等方面明显优于桂枝汤。可见黄芪桂枝五物汤与气虚冻伤模型的相关程度较桂枝汤更高。

Objective：To establish a model of qi deficiency-frostbite rats and to explore the effects and mechanism of Huangqi Guizhiwuwu Decoction（HWD）and Guizhi Decoction（GZD）,and to evaluate the relativity to the model.Methods：The model of qi deficiency-frostbite rats was prepared by the method of overstrain and irregular diet combined with soaking feet in hypothermia ethanol-water mixture.Determining the levels of some cytokine and the contents of TXB2 and PGF1αin serum of rats treated with HWD and GZD by enzyme-linked immunosorbent assay method.Results：IL-1,IL-2,IL-6,TNF,TXB2,TXB2/PGF1αlevels were significantly higher（P0.01 or P0.001）and PGF1αlevels were lower（P0.001）in serum of rats in the untreated group than those in the normal group.Compared with those in the untreated group,the level of IL-6,TNF,TXB2,TXB2/PGF1αobviously decreased（P0.05,P0.01 or P0.001）and PGF1αobviously increased（P0.01 or P0.001）while that of IL-2 with no significant differences in serum of the rats treated by HWD and GZD;The level of IL-1 obviously decreased （P0.01）in serum of the rats treated by HWD while with no significant differences by GZD;The effects of HWD were much better than those of GZD in regulating the level of IL-6,TXB2,TXB2/PGF1α（P0.05）.Conclusion：The model of qi deficiency- frostbite of rats has inflammatory response,immunologic dysfunction and a state of forming thrombus easily.The HWD and GZD can have effect on inhibiting immunological stress,improving blood circulation and pathological injury.But HWD played a better role in improving the pathological injury,inflammatory response and inhibiting platelet aggregation than GZD.It can be seen that the degree of correlation of HWD with the model of qi deficiency-frostbite is higher than that of GZD.
