摘要
为弄清IGF-I在山羊卵泡不同发育阶段对颗粒细胞增殖与分化的影响,分类采集山羊小腔(1~3mm)和大腔(5~8mm)卵泡颗粒细胞进行体外培养,对培养后颗粒细胞的DNA含量和孕酮分泌量进行测定。结果表明,IGF-I对来源于大、小卵泡颗粒细胞的增殖与分化都有促进作用,对小卵泡颗粒细胞的促增殖作用强,促分化作用弱,而对大卵泡颗粒细胞的促分化作用强,促增殖作用弱。这种差异很可能由大、小卵泡颗粒细胞的增殖与分化活性不同引起。
To study the effects of IGF-I on proliferation and differentiation of
granulosa cells during the development of antral follicles in goat, granulosa cells from small
antral follicles (1 ̄3 mm in demeter) and large antral follicles (5 ̄8 mm in demeter) were
cultured in vitro respectively, the granulosa cell DNA content and progesterone secreation were
measured at the end of culture. It is showed that, granulosa cells from small antral follicles with
an intrinsic high proliferative activity and a low differentiated capbility and granulosa cells from
large antral follicles with a restricted proliferative activity and a high differentiated capbility.
IGF-I promote both proliferative and differentiated activity in granulosa cells from both large
and small antral follicles, but the proliferationpromoting effect in cells from small antral
follicles is higher and in cells from large follicles is lower, on the contrary,the
differentiationpromoting effect is higher in cells from large follicles and lower in cells from small
follicles,this difference effects could be related to difference proliferation and differentiation
activity of cells from small and large follicles.
