摘要
目的:观察人富血小板血浆(PRP)及其细化成分对小鼠成骨细胞MC3T3-E1增殖的影响。方法:抽取年轻男性志愿者静脉血制备富血小板血浆,并提取洗涤血小板(WPLT)和乏血小板血浆(PPP)等细化成分,使用细胞定量测定试剂盒测定细胞增殖情况。结果:50ml/L的PRP对MC3T3-E1细胞的增殖没有明显影响,而50ml/L的WPLT显著促进了MC3T3-E1细胞的增殖,但是,这种促进增殖的作用效果会随着WPLT浓度的倍比增加而显著降低,并且当其浓度为150ml/L时反而会显著抑制细胞的增殖。另外,50ml/L的PPP对MC3T3-E1细胞的增殖有明显的抑制作用,而且倍比添加PPP会显著抑制50ml/LWPLT促进的MC3T3-E1细胞增殖。结论:人WPLT促进小鼠成骨细胞MC3T3-E1增殖的作用具有浓度特异性,PPP对MC3T3-E1的增殖有显著的抑制作用。
Objective:To investigate the effects of different components in human platelet-rich plasma(PRP)on the proliferation of mouse osteoblast MC3T3-E1 cells.Methods:PRP was obtained from venous blood samples of a young man volunteer.Washed platelet(WPLT)and platelet-poor plasma(PPP)were extracted from the PRP.Cell quantification kit was applied to detect cell proliferation.Results:50 ml/L PRP had no significant effects on MC3T3-E1 cell proliferation,while 50 ml/L WPLT apparently promoted the proliferation of MC3T3-E1 cells.However this promoting effect decreased with the WPLT concentration increased.WPLT at 150 mol/L inhibited the MC3T3-E1 cell proliferation.In addition,50 ml/L PPP significantly inhibited cell proliferation,and PPP inhibited MC3T3-E1 cell proliferation promoted by 50 ml/L WPLT in a dose dependent manner.Conclusion:Human WPLT has a proliferation promoting effect on MC3T3-E1 cells.However,PPP may inhibit MC3T3-E1 cells proliferation.
