电离辐射对人肝细胞子代克隆热休克蛋白HSP60表达的影响

摘要 采用激光共聚焦显微技术和Western blot技术观察热休克蛋白60(HSP60)在受照人肝细胞克隆子代中的表达情况,并对其在不同受照剂量组中的表达量进行分析,探讨辐射对人肝细胞克隆子代HSP60蛋白表达的影响。结果显示HSP60蛋白主要集中于细胞核的周围,荧光定量分析结果表明HSP60蛋白表达量随着照射剂量的增加出现上调趋势。Western blot测定结果显示各剂量照射组克隆子代中HSP60蛋白随照射剂量的增加而表达增强,该结果与激光共聚焦显微观察结果一致。上述结果表明辐射可使受照人肝细胞子代中的HSP60蛋白表达发生改变,该结果可为探讨辐射诱发基因组不稳定性潜在的分子机理提供基础资料。 This study was designed to characterize the expression of HSP60 in progeny of human liver cells irradiated by γ ray, Laser Scanning Confocal Microscopy (ISCM) and Western blot were used to detect HSP60 in progeny of normal human liver ceils exposed to 2, 4 or 6 Gy γ ray from a 60 Co unit at a dose rate of 0.53 Gy/min. The results demonstrated that HSP60 localized in the cytoplasm of progeny of human liver cells. LSCM and Western blot both showed that the expression level of HSP60 was up-regulated in progeny of irradiated human liver cells in a dose-dependent manner. These results can provide the basis for study of genomic instability induced by radiation at the molecular level.
