摘要
目的:研究血脉通胶囊(XMT)对急性心肌缺血MMP-9和NF-κB表达的影响,为深入探讨血脉通胶囊对心肌缺血的保护机制提供实验依据。方法:60只大鼠随机分为5组,正常对照组、模型组、血脉通胶囊高、中、低剂量组。各给药组连续灌胃给药14 d后,除对照组外,其余各组均采用皮下注射异丙肾上腺素(ISO),每次30 mg.kg-1,每日1次,连续2 d,诱导急性心肌缺血模型。分别记录注射前、注射后5,10,20,30 min时心电图变化;HE染色观察心肌病理组织学改变,免疫组化及RT-PCR法观察MMP-9,NF-κB在心肌组织中蛋白质及mRNA的表达。结果:血脉通胶囊高、中、低剂量组和模型组相比,可不同程度降低心肌缺血心电图ΔΣ-ST水平;改善心肌缺血的病理组织学改变;血脉通胶囊高剂量组可减少心肌MMP-9,NF-κB蛋白及mRNA表达(P<0.05)。结论:血脉通胶囊对ISO所致大鼠急性心肌缺血有一定的预防作用,其机制可能与血脉通胶囊降低心肌组织MMP-9,NF-κB的表达有关。
Objective: To investigate the expression change of matrix metalloproteinase-9(MMP-9) and nuclear factor-κB(NF-κB) induced by Xuemaitong capsule(XMT) preconditioning on rats of myocardial ischemia,and provide experimental evidence for the protection of XMT.Method: 60 wistar rats were randomly divided into 5 groups: the control group,the model group,the three difference dose groups of XMT.The groups of XMT were treated by continuous 14 days intragastric administration.After 14 days,except the control group,all groups were continuous 2 days subcutaneous injected with isoproterenol 30 mg·kg-1 every day.Before and 5,10,20,30 minutes after subcutaneous injection,electrocardiogram were recorded,immunohistochemical staining,RT-PCR and image analysis were also measured for the investigation of the expression of MMP-9 and NF-κB in the ischemic region.Result: Compared with the model group,the ΔΣ-ST of electrocardiogram of the high dose,middle dose,low dose groups of XMT decreased at different degree.Pathological conditions of ischemic myocardial ameliorated.The immunohistochemical staining and RT-PCR indicated that the expression of MMP-9 and NF-κB decreased in the high dose group(P0.05),the mRNA expression of MMP-9 and NF-κB decreased in the high dose group(P0.05).Conclusion: The expression of MMP-9 and NF-κB increased after myocardial ischemia.After high dose treated by XMT,the expression reduction of MMP-9 and NF-κB indicated that XMT played an important preventive role in acute myocardial ischemia.
