北冬虫夏草对一次力竭小鼠血淋巴细胞DNA损伤的影响 被引量:2

Effect of Cordyceps Militaris L on DNA Damage of Lymphocyte in Blood of Mice After Exhaustive Exercise

摘要 目的:探讨北冬虫夏草对力竭性运动后机体自由基代谢的影响和消除运动性疲劳的机理。方法:昆明种小鼠64只,随机分为四组:正常对照组(NC组)16只、运动对照组(EC组)16只、运动+北冬虫夏草组(ECM组)16只、运动+人参组(EP组)16只。采用单细胞凝胶电泳法、硫代巴比妥酸法、黄嘌呤氧化酶法和二硫代二硝基苯甲酸法分别测定血液淋巴细胞DNA的损伤、血清MDA的含量、血清SOD和GSH-Px的活性。结果:①与NC组相比,EC组大鼠血清MDA含量显著增加(P<0.01),血清SOD和GSH-Px活性显著下降(P<0.01)。与EC组相比,ECM组大鼠血清MDA含量显著下降(P<0.01),血清SOD活性明显增高(P<0.05),GSH-Px活性显著增高(P<0.01);EP组大鼠血清MDA含量显著下降(P<0.01),血清SOD和GSH-Px活性显著增高(P<0.01)。②与NC组相比,EC组大鼠血液淋巴细胞尾长、尾矩及椭圆矩显著增加(P<0.01)。与EC组相比,ECM组大鼠血液淋巴细胞尾长、尾矩及椭圆矩显著降低(P<0.01);EP组大鼠血液淋巴细胞尾长、尾矩及椭圆矩显著降低(P<0.01)。结论:北冬虫夏草可以提高小鼠血清SOD、GSH-Px的活性,降低血清MDA的含量,自由基对DNA的直接损伤,对一次力竭运动导致的小鼠血液淋巴细胞DNA的损伤具有保护作用,与人参的作用效果相类似。 Objective: To provide the experimental evidences for Cordyceps militaris L staving and eliminating exercise-induced fatigue. Methods: sixty mouses were divided into 4 groups at random: normal contrast group ( NC, n = 16 ), exercise contrast group( EC, n = 16 ), exercise and Cordyceps militaris L group (ECM, n = 16 ), exercise and Panax ginseng group (EP,n = 16). The authors adopted exhaustive exercise. In order to make sure there were 8 rats in every group,the authors eliminated the superabundance rats randomly after the exercise and measured the DNA damage of lymphocyte, malondialdehyde (MDA) content,superoxide dismutase (SOD) and glutathione peroxidase (GSH-Px) activities in serum of rats by SCGE, TBA, XO and DTNB methods. Results: ( 1 ) Compared with NC group, the MDA content increased remarkably ( P 〈 0.01 ), the SOD and GSH-Px activities in serum of rats decreased markedly in EC group ( P 〈 0. 01 ). Compared with EC group, the MDA content decreased remarkably ( P 〈 0. 01 ), the SOD activities in serum of rats increased in ECM group ( P 〈 0.05 ), the GSH-Px activities in serum of rats increased markedly in ECM group ( P 〈 0. 01 ) ; the MDA content decreased markedly ( P 〈 0.01 ), the SOD and GSH-Px activities in serum of rats increased remarkably in EP group rats ( P 〈 0. 01 ). (2)Compared with NC group,Tail length,Tail moment and Olive moment of lymphocyte increased observably in EC group rats( P 〈0.01 ). Compared with EC group ,Tail length,Tail moment and Olive moment in ECM group rats reduced evidently( P 〈 0.01 ) ;there were an obvious decrease in Tail length, Tail moment and Olive moment (P 〈 0. 01 )in EP group rats. Conclusion:( 1 ) Cordyceps militaris L can eliminate the free radical, enhance the SOD and GSH-Px activities in serum of rats and reduce the damage of DNA caused by OH · etc. (2) Cordyceps militaris L can attenuate exerciseinduced DNA damage after exhaustive exercise. The function is similar to that of Panax ginseng.
