巴西橡胶树SUMO活化酶基因HbSAE1的克隆及表达 被引量:2

摘要 根据一个从巴西橡胶树胶乳cDNA文库中获得的EST片段的序列设计引物,通过RACE的方法获得了橡胶树编码SUMO活化酶(SUMO-activating enzyme,SAE)的cDNA,命名为HbSAE1。序列分析结果表明HbSAE1长为2 411 bp,含有1917 bp的阅读框,214 bp的5'-UTR和280 bp的3'-UTR,编码638个氨基酸,分子量为70.79 ku,等电点为5.41。半定量RT-PCR分析结果表明HbSAE1基因在花、树皮、叶、胶乳中均有表达,其中在胶乳中表达量最高。 The full-length cDNA encoding SUMO-activating enzyme,designated HbSAE1,was isolated from rubber tree based on the EST cloned from a cDNA library of latex.Sequence analysis showed that the length of HbSAE1 is 2 411 bp,it consisted of a 945 bp open reading frame,a 214 bp 5’-UTR and a 280 bp 3’-UTR,as well as encoded 638 amino acids with molecular weight of 70.79 ku,isoelectric point(pI of 5.41.The deduced amino acid sequences of HbSAE1 showed high identities of 79%,72% and 55% to those of the SUMO-activating enzyme protein from Ricinus communis,Selaginella moellendorffii and Arabidopsis thaliana.The results of RT-PCR analysis indicated that HbSAE1 transcripts accumulated at relatively low levels in the flower,leaves and bark,while HbSAE1 transcripts accumulated at relatively high levels in the latex.
