摘要
目前高温平菇菌种存在混乱。本研究对16个供试菌株进行ITS特异性扩增,得到约680 bp的单一片段。根据遗传差异选择菌株进行ITS克隆测序,经ITS序列和ITS-RFLP分析可知:16个菌株分为二个种类,第一类由Pl.cy0002、Pl.cy0009、Pl.cy0012、Pl.cy0014和Pl.cy0015等5个菌株组成,为肺形侧耳;第二类是由剩下的11个菌株组成,是糙皮侧耳类,其中菌株Pl.cy0001和Pl.cy0006的ITS序列与其它菌株存在差异。研究结果表明基于ITS序列建立的ITS-RFLP可应用于高温平菇种的鉴定。
At present,the identification of the strains of thermotolerant Pleurotus spp.were confusing.In this study,internal transcribed spacer(ITS) of sixteen strains of thermotolerant Pleurotus spp.were specially amplified,and a single 680 bp product was gotten.The sixteen strains were clustered into two groups based on ITS-RFLP and ITS sequence analyses.The first group,which belonged to Pleurotus pulmonarius,included Pl.cy0002,Pl.cy0009,Pl.cy0012,Pl.cy0014 and Pl.cy0015.The rest of the strains belonged to the second group,Pleurotus ostreatus.In addition,the ITS sequences of Pl.cy0001and Pl.cy0006 were different from other strains in the second group observed.The results showed that ITS-RFLP based on the ITS sequence could be applied to the identification of thermotolerant Pleurotus spp.species.
