摘要
利用吸附法,将圆柱状假丝酵母脂肪酶固定于4种疏水性不同的载体上.固定化酶的活性及稳定性随载体疏水性的增大而增大.用YWG-C6H5作为载体,在有机溶剂-水双液相体系中催化萘普生甲酯的不对称水解,反应120h转化率为24.72%,产品的对映体过量值(eep)为94.82%.并发现添加乳糖有助于提高固定化酶的稳定性,固定化酶批式连续反应半衰期达1000h以上.
In this paper the properties of Candida rugosa lipase immobilized on four kinds of support by an absorption method in organic\|aqueous biphase system have been studied. It was found that the immobilized lipase in organic media remained a very high activity, which was highly dependent on the nature of support material. The immobilized lipase activity in organic\|aqueous biphase media decreased with increasing hydrophilicity of the support materials. With YWG\|C6H5 as the support for lipase immobilization, 94.82% enantiomeric excess of naproxen could be obtain with a 24.72% conversion of naproxen methyl ester. The lactose addition showed an improvement in immobilized enzyme stability, while the half\|life period of immobilized enzyme was more than 1000 h.
