清化消瘀方对糖尿病大鼠局灶性脑缺血再灌注后NF-κBp65、ICAM-1表达的影响

摘要 目的:观察清化消瘀方对糖尿病局灶性脑缺血再灌注损伤大鼠核转录因子(NF-κBp65)、细胞间粘附因子(ICAM-1)表达的影响。方法:将大鼠随机分为空白对照组、假手术组、局灶性脑缺血再灌注组、糖尿病局灶性脑缺血再灌注组、清化消瘀方组,采用链脲佐菌素(STZ)和线栓法建立糖尿病大鼠大脑中动脉闭塞再灌注模型,应用免疫组化法检测NF-κBp65、ICAM-1表达水平。结果:空白对照组、假手术组脑组织NF-κBp65表达微弱,与假手术组相比,脑缺血再灌注组、糖尿病脑缺血再灌注组、清化消瘀方组的NF-κBp65的阳性表达增加(P<0.01);清化消瘀方组NF-κBp65表达低于脑缺血再灌注组、糖尿病脑缺血再灌注组(P<0.05);空白对照组、假手术组未见ICAM-1阳性表达。脑缺血再灌注组、糖尿病脑缺血再灌注组表达增加,清化消瘀方组ICAM-1免疫阳性血管数较脑缺血再灌注组、糖尿病脑缺血再灌注组著减少(P<0.05)。结论:清化消瘀方能够降低糖尿病脑缺血再灌注损伤大鼠NF-κBp65、ICAM-1的表达,对糖尿病并发脑卒中有治疗作用。 Objective:To investigate the mechanisms of Qinghuaxiaoyu Decoction on the Expression of NF-κB,ICAM-1 in diabetic rats with Focal Cerebral Ischemia-Reperfusion Injury.Methods:The rats were randomly divided into five groups:normal group,the sham-operation group,the MCAO(Middle Cerebral Artery Occlusion) group,the diabetic cerebral ischemic reperfusion group and herbal-decocted diabetic cerebral ischemic reperfusion group.The rats were respectively divided into three subsets:reperfusion for 6h,12h and24h(n =8).Diabetic model was made by injection of streptozocin(STZ) through abdomen.The model of cerebral ischemia reperfusion was developed by thread embolism method.Immunohistochemical method was performed to examine the expression of NF-κB and ICAM-1 method at different time point after reperfusion.Results:The exprssion of NF-κBp65 and ICAM-1 with the treatment of Qinghuaxiaoyu Decoction are decreased in Treatment group,there are significant differences(P〈0.01 or P〈0.05).Conclusions:Qinghuaxiaoyu Decoction decreased the expression of NF-κB and ICAM-1 in diabetic rats with Focal Cerebral Ischemia-Reperfusion Injury which maybe one of the effective curative mechanisms for the diabetes merged with stroke.
