摘要
目的:探讨乌拉地尔对缺氧复氧的血管内皮细胞内皮素(ET)-1分泌和释放以及ET-1mRNA表达的影响。方法:培养人脐静脉内皮细胞,制作缺氧复氧模型。分为对照组、缺氧复氧组、缺氧复氧+乌拉地尔1、5、20、100μmmolL组。用逆转录聚合酶链反应方法半定量研究人脐静脉内皮细胞ET-1mRNA的表达;用酶联免疫吸附双抗体夹心法检测各组细胞上清液中ET-1的含量。结果:(1)缺氧复氧的人脐静脉内皮细胞,ET-1产生增加,随时间延长增加明显。乌拉地尔处理后,ET-1水平增加幅度逐渐降低,呈浓度依赖性和时间依赖性(P均<0.05)。(2)缺氧复氧的人脐静脉内皮细胞,ET-1mRNA表达增加,乌拉地尔处理后,ET-1mRNA表达减少(P均<0.05)。结论:乌拉地尔减少了缺氧复氧人脐静脉内皮细胞ET-1分泌和释放,减少ET-1mRNA的表达。
Objective To investigate the effects of urapidil on the expressions of endothelin-1 and its mRNA in hypooxide-reoxide human umbilical vein endothelial cells. Methods Hypooxide-reoxide models, which were made from human umbilical vein endothelial cells, were divided into control group, hypooxide- reoxide group, hypooxide-reoxide and urapidil (1, 5, 20, 100 μmol/L) groups. The expression of ET-1 mRNA was measured by reverse transcription polymerase chain reaction and that of ET-1 was by enzyme- linked immunosorhent assay. Results ET-1 level was increased with time increasing in hypooxide-reoxide human umbilical vein endothelial cells (P 〈 0.05). After treated with urapidil, the increased ET-1 level was decreased with time and concentration of urapidil increasing (P 〈 0.05). The expression of ET-t mRNA was increased (P 〈 0.05), but after treated with urapidil, it was decreased (P 〈 0.05). Conclusions Urapidil could decrease the expressions of ET-1 and its mRNA in hypooxide-reoxide human umbilical vein endothelial cells.
