摘要
目的:检测Meynert核团(Nucless basalis of meynert,nbM)神经细胞膜上钠、钾、钙通道电流。方法:采用细胞急性分离技术获得单个神经元,运用单细胞膜片钳技术全细胞记录方式记录钠、钾、钙通道电流。结果:钠电流激活失活均很迅速-,50 mV时峰值电流达到最大,约3~4 nA;钾离子电流分为瞬时外向钾电流(IA)和延迟整流性钾电流(IK)两部分I,A部分失活较快I,k部分几乎不失活,峰值电流最大约为7~8 nA;记录出内向的钙电流,失活较慢,大约0~10 mV时峰值电流达到最大,通过运用CdCl2证明该电流为Ca2+离子所介导的高电位激活性钙电流。结论:在nbM神经元上能够诱导出钠、钾、钙电压门控通道电流,为进一步相关研究提供了实验依据。
Objective:Torecord normal sodiumcurrent,potassiumcurrent,and calciumcurrent on neurons ofnucleus basalis of Meynert (nbM). Methods:Single-cell was obtained with cell acutely dissociated technique,sodium current,potassium current,and calcium current were recorded by patch-clamp technique of single cell under whole cell patch pattern. Results:The activation and inactivation of sodium current were showed swiftly. The sodium peak current reached maximum,about 3~4 nA at-50 mV. Potassium current included transient outward potassium currents (I A )and delayed rectifier-like potassium currents (I k ). I A was activated swiftly,I k was hardly inactivated. The Potassium peak current was about 7~8 nA. The inward calcium current was inactivated slowly. The calcium peak current reached maximum ,whilethevoltagewas0~10 mV.Thecurrentwasblocked by20 μmol/LCdCl 2 completely. Conclusion:The voltage-gated sodium channel current ,voltage-gated potassium channel current,and voltage-gated calcium channel current were recorded successfully. It provided experimental data for further research.
