5-氟脲嘧啶联合尼美舒利对人食管癌EC109细胞VEGF表达的研究

摘要 目的:通过观察5-氟脲嘧啶(5-Fluorouracil,5-FU)、尼美舒利(Nimesulide,NIM)单独或联合作用对人食管癌EC109细胞血管内皮生长因子(Vascular endothelial growth factor,VEGF)表达的影响,初步探讨5-FU、NIM抑制食管癌血管生成的机制。方法:培养食管癌EC109细胞,不同浓度5-FU、NIM单独或联合作用24 h后,MTT法检测细胞增殖情况;流式细胞仪检测细胞凋亡;RT-PCR检测VEGF mRNA表达情况;Western blot法检测VEGF蛋白表达。结果:NIM与5-FU联合作用于食管癌EC109细胞株时,其抑制率与凋亡率较两者单独作用时高,且VEGF mRNA及蛋白的表达水平低于两者单独作用时的表达水平。结论:NIM联合5-FU能有效抑制食管癌EC109细胞增殖,其中抑制VEGF表达可能为其机制之一。 Objective:To investigate the effects ofNIM(a cyclooxygenase 2 inhibitor),5-FU,and their combination on the expression of VEGF,in order to explore the possible mechanism. Methods:Nimesulide (100,200,400μmol/ml)and 5-FU(100,200,400μmol/ml) were used.EC109 cell lines were divided into control group,Nimesulide treated group,5-FU treated group,and Nimesulide combined with 5-FU group.The inhibition on the proliferation of EC109 cells respectively treated with Nimesulide ,5-FU,and their combination were detected by MTT and their apoptosis rates were detected by FCM.The expression of VEGF mRNA was detected by RT-PCR. The protein expression of VEGF was detected by Western blotting. Results :The inhibition rate and apoptosis rate ofNimesulide combined with 5-FU were significantly higher than the single Nimesulide or 5-FU single treated.Nimesulide combined with 5-FU significantly reduced VEGF mRNA and protein ,as compared with the single Nimesulide and 5-Fu.Conclusion:Nimesulide combined with 5-FU can inhibit the EC109 esophageal cancer cell proliferation in vitro,inhibiting VEGF expression may be one of its mechanisms.
