罗格列酮联合还原型谷胱甘肽对非酒精性脂肪肝大鼠胰岛素抵抗作用的研究 被引量:1

摘要 目的研究罗格列酮联合还原型谷胱甘肽对非酒精性脂肪肝(NAFL)大鼠胰岛素抵抗状态及相关生化指标的影响。方法建立NAFL大鼠模型。设计正常组、模型组、罗格列酮组、罗格列酮联合还原型谷胱甘肽组,应用ELISA、免疫组化等方法检测血清和胰腺内胰岛素水平,同时检测血清各生化指标。结果模型组血清白蛋白、ALT、总胆固醇显著高于正常组(P<0.01),甘油三酯、空腹血糖水平显著低于正常组(P<0.01);罗格列酮对上述少数生化指标有改善作用,但与模型组相比无显著差异(P>0.05);模型组血清和胰腺组织内胰岛素水平显著高于正常组(P<0.01),而罗格列酮组血清和胰腺组织内胰岛素水平较模型组显著下降(P<0.01);模型组血清胰岛素抵抗指数明显高于正常组(P<0.01),罗格列酮单独或联合还原型谷胱甘肽组胰岛素抵抗指数明显下降(P<0.01)。结论大多数NAFL大鼠肝功能指标出现异常,同时出现明显的胰岛素抵抗,罗格列酮可以不同程度改善部分生化指标,对其胰岛素抵抗状态具有显著改善作用,但罗格列酮和还原型谷胱甘肽没有显著的联合作用。 Objective To investigate the effects of rosiglitazone combined with reduced glutathione on insulin resistance and biochemical related indicators in rats nonalcoholic fatty liver disease ( NAFL) . Methods NAFL rats models were produced. Rats were divided into four groups: normal group,model group,rosiglitazone group,group of rosiglitazone combined with reduced glutathione. Insulin in serum and pancreas were detected by immunohistochemisty and ELASA,and the biochemical indicators were detected at the same time. Content or intensity of insulin were compared among the above groups. Results The levels of albumin,ALT,total cholesterol in model group increased obviously compared with normal group ( P 0. 01) ,the concentration of triglyceride and fasting blood glucose in model group decreased markedly ( P 0. 01) . Rosiglitazone could reverse the levels of biochemical indicators compared with model group,but there was no significant difference. The levels of insulin in serum and pancreatic tissue in model group were obviously higher than those in normal group ( P 0. 01) . Rosiglitazone could reduce the level of insulin obviously ( P 0. 01) . The changing trend of insulin resistance index in above four groups were similar with insulin. Conclusion Some biochemical indicators in NAFL rats showed abnormity,and the insulin resistance occurred obviously compared with healthy rats. The rosiglitazone can reverse the levels of some biochemical indicators and decrease the insulin resistance index in NAFL rats,but combined treatment occurs no cooperative effects.
