摘要
目的:探讨prostasin基因在乳腺癌及相应癌旁组织中的表达,并探讨prostasin与乳腺癌临床病理因素的关系。方法:用RT-PCR法检测prostasin在40对包括原发乳腺癌组织及相应癌旁组织共80例组织中的表达,其中24例乳腺癌伴有转移,余16例不伴有转移。结果:40例乳腺癌组织中,prostasin表达阳性12例(30%),40例相应乳腺癌旁组织中prostasin表达阳性31例(77.5%),两者相比差别有显著统计学意义(P<0.01)。乳腺癌中prostasin低表达与乳腺癌的分期、有无转移有相关性(P<0.05),而与患者年龄、肿瘤大小、雌激素受体(ER)、孕激素受体(PR)、C-erbB-2、Ki-67无相关性。结论:prostasin基因在乳腺癌组织中低表达,可能是导致乳腺癌发生发展的一种抑癌基因并可能成为乳腺癌治疗的新靶点。
Objective:To investigate the expression of prostasin in breast carcinoma and explore its relationship with clinicopathological factors of breast cancer.Methods:Prostasin mRNA expression was detected by reverse transcription-polymerase chain reaction(RT-PCR) in 40 cases of primary breast cancer tissues and adjacent breast tissues.In 40 breast cancer tissues,there were 24 cases with metastasis and the other 16 cases without metastasis.Results:The expression rates of prostasin mRNA in breast cancer and adjacent tissues were 30% and 77.5% respectively.Expression of prostasin was lower in breast cancer tissues than that in adjacent tissues(P 0.01).Underexpression of prostasin in breast cancer was correlated with lymph node metastasis,clinical stage(P 0.05).No correlation was found between the expression of prostasin and age,tumor size,ER,PR,C-erbB-2(P 0.05).Conclusion:Prostasin is lowly expressed in breast cancer,prostasin gene maybe is an anti-oncogene in the development of breast cancer and may become a new therapeutic target.
