摘要
目的探讨三氯乙烯(TCE)对豚鼠胸腺淋巴细胞凋亡基因BAX、BAD和Bcl-2 mRNA表达的影响。方法采用豚鼠最大值法(GPMT),将豚鼠随机分为3组,分别为阴性对照组、阳性对照组、TCE实验组,用皮内注射的方式分别注射橄榄油、2,4-二硝基氯苯(DNCB)和三氯乙烯(TCE),实验结束后取豚鼠胸腺淋巴细胞,用荧光定量PCR检测凋亡基因表达水平。结果 阳性对照组和TCE实验组动物出现明显皮肤损害。阳性对照组BAX和BAD mRNA表达水平比阴性对照组分别高158%和136%,差异有统计学意义(P<0.01);TCE实验组BAX和BAD mRNA表达水平比阴性对照组分别高86%和150%,差异有统计学意义(P<0.01);但阳性对照组和TCE实验组Bcl-2 mRNA表达无明显改变。结论三氯乙烯可诱导豚鼠产生明显的皮肤变态反应,对豚鼠胸腺淋巴细胞凋亡基因表达水平有一定影响。
Objective To study mRNA expressions of apoptosis genes BAX、BAD、Bcl-2 in thymus gland of guinea pigs after treatment with trichloroethylene(TCE).Methods Guinea pig maximization test(GPMT) was applied in this study,guinea pigs were divided into 3 groups,namely negative control,positive control and TCE treatment.Animals of 3 groups were administrated with olive oil,2,4-dinitrochlorobenzene(DNCB),and TCE,respectively,by intradermal injection.The thymus glands were taken out after various treatment,mRNA expressions of apoptosis genes BAX、BAD、Bcl-2 in thymus gland of guinea pigs were detected by real-time fluorescent PCR assay.Results Obvious skin impairment was observed in the groups of positive control and TCE treatment,the skin impairment included erythema and edema.The levels of BAX and BAD mRNA expression increased by 158% and 136% in TCE treatment group when compared with control,BAX and BAD mRNA expression levels in group of positive control increased by 86% and 150% when compared with negative control.However,the level of Bcl-2 mRNA expression did not elevate significantly when compared with negative control.Conclusion Trichloroethylene could induce skin allergic reaction in guinea pigs,it also brought about obvious increase in mRNA expression of apoptosis genes BAX、BAD in thymus gland.
