壬基酚对大鼠胰腺内分泌功能的影响 被引量:6

摘要 目的研究不同剂量壬基酚(NP)对大鼠胰腺内分泌功能的影响,探讨内分泌干扰物NP对大鼠胰腺的毒性作用。方法健康4周龄SD大鼠40只,随机分为溶剂对照组、低剂量组(40 mg/kg)、中剂量组(80 mg/kg)、高剂量组(200 mg/kg)共4组,每组有大鼠10只,雌雄各半,灌胃染毒。染毒15 d后观察其血糖的浓度、用放射免疫法测定血浆胰岛素和胰高血糖素的变化,免疫组织化学观察胰腺胰岛损伤程度。结果 (1)染毒15 d后,各实验组血糖、血浆胰高血糖素浓度呈逐渐增高的趋势,与对照组相比差别有统计学意义(P<0.05);(2)各实验组的血浆胰岛素含量呈逐渐降低趋势,与对照组相比差别有统计学意义(P<0.05);(3)免疫组化结果显示各实验组大鼠胰岛变小,仅有少数弱阳性反应的β细胞,提示β细胞严重损伤,中剂量组大鼠胰岛阳性β细胞染色比高剂量组弱,但比低剂量组强。各实验组阳性α细胞暗褐色染色数量较多呈宽带状分布。结论 NP可损伤胰腺胰岛β细胞,可刺激胰岛α细胞增生,并且血糖浓度明显增高;同时NP可造成胰腺内分泌功能减低。
