Tissue factor (TF, coagulation factor Ⅲ ) is distributed widely in extravascular tissues.It is a transmembrane glycoprotein located on the surface of a number of cell types and serves as an essential cofactor for factor Ⅶa. Blood cells and endothclial cells normally do not express TF. Various pathogenic agents can cause the dysfunction of endothelial cells and induce TF synthesis. The binding of plasma factor F Ⅶ /Ⅶ a to TF located on the surface of the cell may initiate activation of coagulation. The abnormal expression and release of TF in endothelial cells and subendothelial cells result from dysfunction of the endothelium can lead to the disorder of the coagulant fuction in the disease state. Thus, some cardiovascular disease associated with the hypercoagulant state and thrombosis will occur. In addition, TF, as a highaffinity receptor, participates in other cellular processes including intracellular signalling and tumor-associated angiogenesis. This brief review attcmpts to summarize present knowledge on the structure and fuction, in vivo and in vitro expression and release of TF and the gene regulation as well as the role thereof in the cardiovascular disease.
