摘要 为了探讨超抗原诱导的淋巴细胞凋亡机制.本实验采用超抗原金黄色葡萄球菌肠毒素B(SEB)处理人外周血单个核细胞,采用流式细胞术检测细胞DNA断裂的百分率;同时动态观察了细胞表面APO-1、FasL的表达及细胞内Bcl-2蛋白的表达。结果表明,SEB不但可引起人外周血淋巴细胞凋亡,而且可诱导淋巴细胞表达APO-1和FasL。细胞凋亡的百分率与细胞表面APO-1和FasL的表达呈正相关(r=0.71,r=0.98),与细胞内Bcl-2表达呈负相关(r=-0.72)。由此可见,APO-1/Fas及其配体参与了超抗原SEB诱导的淋巴细胞凋亡。 To investigate the mechanism of apoptosis of lymphocytes induced by superantigen, human peripheral blood mononuclear lymphocytes were treated with superantigen staphylococcus aureus enterotoxin B (SEB). The percentage of fragmentation of cell DNA was measured by FACS; at the same time, dynamic changes in the expression of APO-1, FasL and Bcl-2 were also studied. The results showed that SEB not only promoted the apoptosis of human peripheral blood lymphocytes,but also induced the expression of APO-1 and FasL. The percentage of apoptotic cells in the SEB group was positively correlated with the expression of APO-1 and FasL (r=0.71; r=0.98), but negatively correlated with the expression of Bc1-2 (r=- 0.72). The results suggest that apoptosis of activated human peripheral blood lymphocytes induced by SEB is closely related to APO-1/Fas death pathway.
