药物诱导大鼠大肠癌组织原卟啉Ⅸ积聚的定量分析 被引量:1

摘要 目的定量分析不同药物诱导大肠癌大鼠大肠正常组织、早期癌和进展期癌组织原卟啉Ⅸ聚积特征。方法15只大鼠分成3组。A组:5-ALA尾静脉注射;B组:DFO腹腔注射半小时后给予5-ALA尾静脉注射;C组:相当量的生理盐水尾静脉注射。每组大鼠诱导4h采集标本。另20只大鼠按B组诱导方法分别在药物诱导2、4、6、8及10h采集标本。用高效液相色谱-荧光检测仪测定其原卟啉Ⅸ含量。结果大肠正常组织、早期大肠癌和进展期大肠癌组织原卟啉Ⅸ含量在A组分别为:(475.261±65.027)、(687.738±73.293)、(852.371±69.457)ng/mg;B组分别为:(496.194±63.539)、(853.194±81.937)、(1173.795±87.184)ng/mg;C组分别为:(347.375±57.413)、(479.375±59.274)、(593.173±37.461)ng/mg。癌变组织原卟啉IX的含量在用药4h最高;正常组织原卟啉IX的含量在用药2h最高,癌变组织与正常大肠组织原卟啉IX含量在用药后4h差异最大。结论5-ALA与DFO联合应用,显著提高5-ALA诱导癌组织原卟啉IX的积聚作用;用药后4~6h检测原卟啉IX含量最高。 【Objectives】The quantitative analysis on the characteristics of accumulation of the protoporphysin Ⅸ of normal tissue,earlier carcinoma tissue,progression carcinoma tissue of colorectal cancer of rat induced by various drugs. 【Methods】15 SD rats with colorectal cancer were divided into three groups. Group A were injected with 5ALA through vena caudalis. Group B were injected by DFO and 5-ALA,first which was injected with DFO through intraperitoneal,after a half hour,injected with 5-ALA through vena caudalis. Group C were injected with isotonic Nacl chloride through vena caudalis. Samples of each group have been gathered after having been induced for 4 hr. Samples of the other 20 rats have been gathered respectively at 2,4,6,8,10 h induced by DFO + 5-ALA. The content of protoporphysin Ⅸ was detected by high performance liquid chromatogram and fluorescence detection. 【Results】5-ALA induced group,the content of protoporphysin Ⅸ of normal tissue,earlier colorectal cancer tissue,progression colorectal cancer tissue respectively acted:(475.261±65.027),(687.738±73.293),(852.371±69.457) ng/ mg; DFO and 5-ALA combine-induced group respectively acted:(496.194±63.539),(853.194±81.937),(1173.795± 87.184) ng/mg. Group C respectively acted:(347.375±57.413),(479.375±59.274),(593.173±37.461) ng/mg. The content of protoporphysin IX of the carcinom tissue achieved tiptop after having been induced for 4 h,which of the normal tissue achieved tiptop after having been induced for 2 hr,which of carcinoma tissue and normal tissue has the most difference after having been induced for 4 hr. 【Conclusions】5-ALA combined with DFO significantly increased the accumulation of protoporphysin Ⅸ in 5-ALA-induced carcinoma. The content of protoporphysin Ⅸ achieved maximum after having been induced by drugs for 4-6 h.
