摘要
目的研究碘化钾(KI)和碘伏(PI)对人皮肤成纤维细胞(HSF)的影响,初步探索碘对HSF的作用机制。方法体外培养HSF,实验分为9组:空白对照组、3mg/LKI组、30mg/LKI组、300mg/LKI组、3000mg/LKI组、1mg/LPI组、2mg/LPI组、3mg/LPI组、4mg/LPI组分别加入不同浓度碘化钾和碘伏作用分别于第2、4、6天采用MTT检测HSF黏附和增殖情况;免疫荧光细胞化学方法观察增殖细胞核抗原(PCNA)表达。结果3mg/LKI组、30mg/LKI组、300mg/LKI组细胞黏附和增殖活性明显增强,PCNA表达增强,与对照组差异有显著性,3000mg/LKI组和含碘伏组细胞活性明显减弱,其中含碘伏组随碘浓度增加细胞活性越差,甚至脱水、死亡。结论适当浓度碘化钾使体外培养HSF黏附和增殖活性增强,高浓度的碘化钾导致细胞凋亡;稀释浓度的碘伏对体外培养HSF有明显毒性作用。
【Objective】To investigate the biological effects of KI and PI on the adhesion and proliferation of skin fibroblasts in vitro and to explore the role of iodine in wound healing.【Methods】Human skin fibroblasts were cultured in vitro,and were divided into 9 groups:control group,3 mg/L KI group,30 mg/L KI group,300 mg/L KI group,3000 mg/L KI group,1 mg/L PI group,2 mg/L PI group,3 mg/L PI group,4 mg/L PI group.After being stimulated by KI and PI with different concentrations,the proliferations of human skin fibroblasts were measured with methyl thiazolyl tetrazolium(MTT) and imm-unofluorescence method respectively.【Results】In 3 mg/L KI group,30 mg/L KI group and 300 mg/L KI group,the adhesion and proliferation of skin fibroblasts were remarkably increased,especially in 3 mg/L group the differences were significant compared with theiodine free control(P〈0.05),but the proliferation activity was progressively retarded in 3000 mg/L concentration.In povidone iodine groups,Fibroblast growth was totally inhibited by povidone iodine solutions.【Conclusions】Proper concentration of KI can promote fi-broblast adhesion and proliferation,but high concentration of KI lead to apoptosis.Even dilute solutions of povidone-iodine were toxic to human fibroblasts.
