摘要
目的以实时荧光定量PCR技术研究BPH与PCa组织标本以及前列腺癌细胞株LNCaP、PC-3中HER-2 mRNA值的表达,探讨HER-2在前列腺癌诊断的特异性意义。方法通过实时荧光定量PCR对23例PCa、37例BPH、31例前列腺癌LNCaP细胞株、22例前列腺癌PC-3细胞株和3例正常前列腺组织HER-2 mRNA的表达,比较其mRNA值定量的差异。结果Pca、LNCaP和PC-3样品HER-2 mRNA表达值与BPH及NT组织差异有显著性(P<0.05)。结论实时荧光RT-PCR定量检测HER-2 mRNA为前列腺癌的诊断、治疗、预后监测等提供了更为可靠的辅助指标。
【Objective】To study the HER-2 mRNA expression in BPH and PCa tissue cells,and LNCaP,PC-3 prostate cancer cells,explore the value of HER-2 mRNA expression level in PCa diagnosis.【Methods】A sensitive,real-time quantitative PCR assay was developed to compare the expression difference of hK2 mRNA in BPH,PCa,LNCaP,PC-3 and normal prostate tissue cells,by testing 23 samples of PCa,37 samples of BHP,37 samples of LNCaP,22 samples of PC-3 and 3 samples of normal prostate tissue.【Results】The expression level ratio of HER-2 in BPH and normal tissues between PCa and HER-2,PC-3 is significantly lower(P〈0.05).【Conclusion】The detection result of HER-2 mRNA expression by RT-PCR can afford more reliable and helpable information for diagnosis,treatment and prognosis of PCa.
